Persimmon (Diospyros Kaki Thunb ‘Saijo’) Peel Improved Dyslipidemia and its Related Production of Atherogenic Autoantigen Complexes in Low- Density Lipoprotein Receptor-Deficient Mice

چکیده
Role of persimmon (Diospyros kaki, Thunb ‘Saijo’) peel was investigated on developing atherosclerosis in low-density lipoprotein receptor (LDLR)-deficient mice with emphasis on lipid metabolism, physio-biological oxidation, production of related atherogenic autoantigens, and production of anti-atherogenic natural antibodies. Male LDLRdeficient mice were fed high fat diet or high fat diet supplemented with 10% dried and powdered persimmon peel (PP) for 12 weeks. PP supplementation significantly prevented the increment of plasma cholesterol and triglyceride levels. High fat diet feeding increased plasma level of oxidized LDL/β2-glycoprotein I (oxLDL/β2GPI) complexes regarded as an atherogenic autoantigen, while PP supplementation significantly blocked the increment (p<0.05). After a 12-week feeding, atherosclerotic plaques in mice fed with diet supplemented with PP decreased by 70% as compared to that of mice fed the high fat diet (p<0.005). PP feeding also reduced urinary 11-dehydro-thromboxane B2, a stable metabolite of the platelet activation marker thromboxane A2, but level of IgM anti-oxLDL antibodies was not changed. Thus, these results obviously demonstrate that persimmon peel may have an anti-atherogenic property through normalization of lipid metabolism and reduced production of the atherogenic complexes.

Data in support of antioxidant activities of the non-extractable fraction of dried persimmon (Diospyros kaki Thunb.)
This data article is related to the research article entitled, "Antioxidant potential in non-extractable fraction of dried persimmon (Diospyros kaki Thunb.)" (Matsumura et al., 2016) [1]. We investigated antioxidant activities of the non-extractable fraction of dried persimmon fruits in vitro and in vivo. We evaluated both extracted fraction and non-extractable fraction, and reported that non-e...

متن کاملEffect of heat treatment on the antioxidative and antigenotoxic activity of extracts from persimmon (Diospyros kaki L.) peel.
Heat treatment of persimmon peel (PP) increased the antioxidative activity of the 70% ethanolic extract (EE) and water extract (WE) from PP. EE and WE both prevented H2O2-induced DNA damage to human peripheral lymphocytes. The antioxidative and antigenotoxic activities of the PP extracts were significantly affected by heating.
متن کاملA Transcriptome Approach Toward Understanding Fruit Softening in Persimmon
Persimmon (Diospyros kaki Thunb.), which is a climacteric fruit, softens in 3-5 weeks after harvest. However, little is known regarding the transcriptional changes that underlie persimmon ripening. In this study, high-throughput de novo RNA sequencing was performed to examine differential expression between freshly harvested (FH) and softened (ST) persimmon fruit peels. Using the Illumina HiSeq...
متن کاملSelection of Suitable Reference Genes for RT-qPCR Normalization under Abiotic Stresses and Hormone Stimulation in Persimmon (Diospyros kaki Thunb)
The success of quantitative real-time reverse transcription polymerase chain reaction (RT-qPCR) to quantify gene expression depends on the stability of the reference genes used for data normalization. To date, systematic screening for reference genes in persimmon (Diospyros kaki Thunb) has never been reported. In this study, 13 candidate reference genes were cloned from 'Nantongxiaofangshi' usi...
